マウスバンク

マウスバンク[1] (英:Mouse Repository)とは、遺伝子改変マウスの作製、収集、保管、供給を行う研究施設である。

遺伝子改変マウスには、外部から特定の遺伝子を導入したトランスジェニックマウス(遺伝子改変動物)や遺伝子を欠損させたノックアウトマウスなどがあり、遺伝子の機能や人間が病気になる原因を調べたり、薬の開発において有効性を調べるモデル動物(ヒト疾患モデルマウス)として利用されている。マウスバンクは、マウスリソースに関する情報や試料を管理することで、生命科学研究における遺伝子改変マウスの利活用を促進し、持続的な研究活動を支える研究基盤になっている。

マウスバンクでは、遺伝子改変マウスを効率的に管理するために生殖工学技術が活用され、凍結保存した受精卵や精子を液体窒素中で保管している。遺伝子改変マウスの受精卵や精子を凍結保存することで、研究者が必要な時に体外受精や胚移植により個体を復元し、速やかに研究に使用することができる。また、遺伝子改変マウスを生体として飼育する必要が無いため、動物実験の基本原則である3Rsにおける動物の削減(英:Reduction)にも貢献している。

マウスバンクは、世界中に創設[2] されており、代表的な研究機関として米国のジャクソン研究所(英:Jackson Laboratory)やカリフォルニア大学デービス校マウスバイオロジープログラム(英:UC Davis Mouse Biology Program[3] )、英国のMRCハーウェル研究所マリー・ライオンセンター(英:Mary Lyon Centre[4] )、日本では熊本大学生命資源研究・支援センター動物資源開発研究施設[5] (Center for Animal Resources and Development, CARD)および理化学研究所バイオリソース研究センター[6] が知られている。

米国におけるマウスバンク組織としてMutant Mouse Regional Resource Centers (MMRRC)[7] 、欧州における マウスバンク組織としてEuropean Mouse Mutant Archive(EMMA)[8] 、アジアにおけるマウスバンク組織としてAsian Mouse Mutagenesis and Resource Association (AMMRA)[9] [10] 、豪州のAustralian Phenomics Facilities(APF)[11] がある。

マウスバンクは国際的に連携しており、各マウスバンクに保管されている遺伝子改変マウスの情報を共有し、International Mouse Strain Resource(IMSR)[12] [13] から検索できるようになっている。研究者は、IMSRで見つけた遺伝子改変マウスをマウスバンクから入手することができる。現在、IMSRでは、80,443系統[14] の遺伝子改変マウスを管理している。

研究者がマウスバンクに寄託した遺伝改変マウスは、マウスに関する遺伝子などの情報がマウスバンクのデータベースに登録され、論文で使用された資源を特定する研究資源識別子であるRRID[15] (英:Research Resource Identifier[16] [17] )が賦与される。RRIDは、研究資源の特定・発見・利用を促進を目的とており、生物リソースを特定するための標準的なフォーマットである。論文に使用した遺伝子改変マウスなどの生物材料のRRIDの引用が、研究の再現性や持続性を向上させる取り組みとしてNature誌などの学術誌で推奨されている[18] 。

医学・生命科学における遺伝子改変動物を用いた研究の再現性、信頼性、完全性を高める国際的な取り組みとして、Improving laboratory animal genetic reporting: LAG-R guidelines(LAG-Rガイドライン)[19] が策定され、マウスバンクにおける試料の保管やIDの管理が推奨されている。
