Erlenmeyer flask

10 grams of beads (2.9 x [10.sup.7] CFU [g.sup.-1]) were added to each Erlenmeyer flask containing 50 [mL.sup.-1] of fermentation medium with initial pH adjusted to 4 and 5, and different sucrose concentrations (15, 20, 25, 30, and 35%).

Afterward, 2ml of each bacterial suspension equivalent to 1 McFarland was inoculated in a 50 mL Erlenmeyer flask containing 20 mL of mineral medium (the composition of the medium in g/L: [(N[H.sub.4]).sub.2] S[O.sub.4] = 5.00; [(N[H.sub.4]).sub.2]HP[O.sub.4] = 1.42;NaCl = 0.50; MgS[O.sub.4].7[H.sub.2]O = 0.40; Ca[Cl.sub.2] = 0.60; KCl = 2.15;FeS[O.sub.4].7H2O = 0.01; ZnS[O.sub.4] = 0.01; CuS[O.sub.4] = 0.01; pH not adjusted) and 1 ml (5%, v/v) of d-limonene.

The cultures were performed in a 250 mL Erlenmeyer flask containing 50 mL medium.

The inoculum medium (100 ml per 250 ml Erlenmeyer flask) was inoculated with 2% inoculum of E.

For analyzing the effect of dye on the growth of yeast isolate YEPD broth medium (100 mL) was prepared and inoculated with yeast culture in 250 mL of Erlenmeyer flask and marked as control.

Analysis of soil enzymes activity: For the analysis of soil urease activity, 5 g soil was taken in an Erlenmeyer flask (100 ml) and 2.5 ml urea solution was added.

An aliquot consisting of 4 g of dried yacon leaves was weighed in an Erlenmeyer flask with a cap, and 100 mL of boiling water was added and allowed to stand for 1 h.

Inoculum medium (35 ml) containing 8 g/L nutrient broth 10 g/L starch 5 g/L lactose and 1.5 g/L NaCl in 50 mM phosphate buffer (pH 7.2) was transferred to a 250 ml Erlenmeyer flask and cotton plugged.

An isolate colony was inoculated into 200 ml of nutrient yeast dextrose broth (NYDB) in 250 ml erlenmeyer flask and incubated at 37 C for 48 hrs.

The Production was carried out in a 250 ml Erlenmeyer flask inoculated with 2ml of broth culture and incubated at 30[degrees]C at 240rpm.
