Brenipatide (INN Tooltip International Nonproprietary Name, USAN Tooltip United States Adopted Name; developmental code name LY-3537031) is a dual agonist of glucagon-like peptide-1 (GLP-1) receptors and gastric inhibitory polypeptide (GIP) receptors.
Brenipatide is under development by Eli Lilly and Company for the treatment of alcoholism, bipolar disorder, asthma, smoking withdrawal, cardiovascular disorders, liver disorders, metabolic disorders, and obesity.[1] [3] [4] [2]
It is taken by subcutaneous injection once per month.[1] [4] [2] The drug has a longer elimination half-life than tirzepatide or retatrutide.[2]
As of December 2025, it was in phase 3 clinical trials for alcoholism and bipolar disorder, phase 2 trials for asthma and smoking withdrawal, and phase 1 trials for cardiovascular disorders, liver disorders, metabolic disorders, and obesity.[1] [4]

Brenipatide is a once-monthly dual-acting peptide from Eli Lilly that targets GLP-1R and GIPR (Table 1) and is currently in a Phase 3 clinical trial for moderate-to-severe alcohol use disorder (NCT07219966). Although it has a single C20 diacid, it exhibits a longer half-life than tirzepatide and retatrutide, likely because of a backbone optimized for stability against enzymatic degradation through a Trp to αMe-Tyr substitution.
